Vlaar gifted Mane and Southampton a second on 13 minutes 46 seconds as his poor back-pass sold Given short to allow Mane to bundle home from close range.
Mane then saved the best for last when he curled Shane Long's pass on the edge of the area past Given to seal his record-breaking hat-trick in fine style on 15 minutes and 18 seconds.
Long then scored twice to extend Southampton's lead to 5-0 before Christian Benteke headed home shortly before half-time to give the shell-shocked Villa fans at least some consolation.
Mane was surprised to hear of his achievement after the match but insisted the result meant more after a run of four league matches without a win.
He told BT Sport: 'It's great. But I think more important for me was the victory and we got that, so I'm happy for the team and the fans.
'We have a chance [to reach Europe] and we'll try to win the next game. It won't be easy but we'll keep going and try to win that next game.
'We have a great team.The last few weeks were not easy but we had to work every day and finally we got the victory.'
Mane's hat-trick, which came after 15 minutes 18 seconds, is also the earliest Premier League hat-trick in history with the previous earliest coming after 22 minutes courtesy of Dwight Yorke in Manchester United's 6-1 demolition of Arsenal in February 2001.
It was the 23-year-old's first hat-trick for the Saints, with the £12m man finding the net after 12 minutes 22 seconds, before adding a second after 13 minutes and 46 seconds, and completing his hat-trick after 15 minutes 18 seconds.
And former record holder Robbie Fowler took to twitter to acknowledge that his record had been beaten, pretending not to know what was happening.
